朝鮮中宗朝之大儒，光祖年十九，遭父喪，事母至孝，師金宏弼，立倡儒學，為士林黨首。
正德十年以學行，承薦擧出仕參奉，
正德十四年官至大司憲，中宗從其議，廢昭閣署，削燕山君廢立以來佐翊之靖國功臣。朝中勛舊咸恨之，南陽君洪景舟之女熙嬪嘗陰使人抹蜜於葉上，作走肖為王狀，引蠶蟲食之，宮人見之，呈於上，中宗意遂不喜，乃照會景舟等，以示除士林之心，
正德十四年十一月十五，洪景舟受詔捕光祖等士林黨八人下獄，幾死，領議政鄭光弼等苦諫不可乃止，流配外道。是年十二月二十日，中宗賜其死，飲鴆毒加縊卒，年三十八，贈首相，諡文正
。中宗既崩，子仁宗立，士林黨皆蒙昭雪，復賢良科，
明穆宗隆慶四年，列之朝鮮四賢。
萬曆三十八年，從祀朝鮮孔廟，其葬龍仁水枝邑，今有墓存焉。
